Abstract
Aeroponic culture by operating with domestic equipment was utilized to produce mycorrhizal fungus inoculum. In the first stage, adzuki bean seeded in pot was adopted as a host plant inoculatcd with Glomus mosseae (GM), After being precolonized, the seedlings of adzuki bean were transplanted in aeroponic culture. The results showed that among the three nutrient solutions used in this study, spore production of mycorrhizal fungus cultivated in 0.5 strength Hoagland’s solution containing 1/4 amount of phosphate was better than that in 0.5 strength Hoagland’s and Taifei nutrient solution (200X). Among the three mycorrhizal fungi used, spore productions of Glomus etunica turn (26 spores / I cm root section) and G. fasclculalum (24 spores) were more abundant than that of G. mosseae (12 spores). In the second stage, tomato, corn and behia grass were used as host plants. The mixture of soil inoculnm of Glomus mosseae,G. etunicatum,G. fasciculatum and G. clarum were inoculated on host plants seedling in pots. After being precolonized by mycorrhizal fungi, seedlings were transplanted in aeroponie culture. The results showed the shoots of corn were too high to be handled with aeroponic culture, and was abandoned in later experiment. Although the spore production per 1 cm colonized root of bahia grass was more abundant than that in tomato. the biomass of colonized tomato roots was higher than that of bahia grass, and tomato has higher economic value, it is worthy to that tomato as a host crop to produce mycorrhizal fungus inoculum in further study. In the third stage, tomato and bahia grass were used as host plants, seeded in spongy lump. After germination, seedlings were transplanted in aeroponic culture. The roots were packaged with a piece of sponge containig mycorrhizae fragments for inoculation. However, the roots near sponge were decaying gradually. Fine net containing mycorrhizae fragment (inoculation net) was adopted to wrap host roots cultivated in aeroponic culture to obtain inoculation, The results showed the development of host root is unaffected, and the infection strenght of mycorrhizal fungi, the production of vesicle and spore were improved. 8 weeks after inoculation, the spore number per 1 cm colonized root of bahia grase is 30, that of tomato is 7.3. For mass production strategy in Taiwan, economic crop such as tomato, could highly improve its sporulation when seedlings were inoculated with mycorrhizal inoculation net after aeroponie culture. The inoculation technique developed in this study is convenient for producer, and therefor suitable for large application and extention.
