Abstract
A modified phenol-LiCl method for the extraction of RNA from seed coats of genotypes L62-1058 and L66-14 (yellow and black seed coated, respectively) of near isogenic lines of soyabean Clark that yielded high quality RNA is described. Seed coat material was ground in a buffer containing a high concentration of bovine serum albumin (100 mg BSA/50 mg of lyophilized seed coats) to competitively inhibit proanthocyanidin binding. The presence of hydrated insoluble polyvinylpolypyrrolidone (PVPP) was also necessary to bind proanthocyanidins and remove them from solution.
