Abstract
RFLP (Restriction Fragment Length Polymorphism) is a relatively new category of genetic marker which allows direct detection of variation at the DNA level. This technology was first developed in 1980 for human genetic study, and has been applied in animal, plant, fungus, and bacteria genetic. RFLP shows some advantages when used in genetic studies, such as co-dominant, simple Mendelian genetic fashion, not tissue or developmental stage specific, and no epistatic interaction. Based on these advantages, it is believed being very useful in woody plant research, and being a powerful tool for enhancing the breeding efficiency in fruit trees. The purpose of this article is to describe some basic techniques in RFLP studies. The potential of using this powerful tool in fruit breeding and some other application have also been discussed.
