台灣地區矮化線蟲Tylenchorhynchus annulatus 及新記錄種T. leviterminalis (Nematoda: Belonolaimidae)之鑑定

Abstract
自2005年1月起，從台彎地區水稻、甘蔗、玉米、香蕉及竹等作物的根圈土中共分離鑑定10群Tylenchorhynchus annulatus (Cassidy, 1930)Golden，1971和9群T. leviterminalis Siddiqi, Mukherjee & DasguPta, 1982，其中後者為台灣地區的新記錄種。各群矮化線蟲之種類鑑定依據外觀形態、形態測量值(morphometrics)及核糖體DNA(ribosoinal DNA，rDNA)片段（包括完整5.8S基因，內轉錄區問ITS-1和ITs-2及部分18S和28S基因）的序列相同度比對分析。本研究提供更多完整的形態測量值（包括b，m，ODG，O，EP、tail及ABW等共7項）和SEM外觀形態方面資料，並首次在GenBank中登錄這二種矮化線蟲的rDNA片段代表性序列，大小皆為1198bp，其序列相同度為91.1%。另二者的ITS-1、ITS-2及5.8S序列相同度（大小）依序分別為88.1%（493bp和490bp）、83.9%（237bp和240bp）及98.8%(162bp)。
Soil samples collected from the rhizosphere of rice, sugarcane, corn, banana, and bamboo in Taiwan yielded 10 populations of Tylenchorhynchus annulatus (Cassidy, 1930) Golden, 1971 and 9 populations of T. leviterminalis Siddiqi, Mukherjee & Dasgupta, 1982., and the latter was a new record in Taiwan. Based on the external morphology, morphometrics and the sequences of ribosomal DNA (including complete 5.8S gene, internal transcribed spacer ITS-1, ITS-2, and partial 18S and 28S gene), each nematode population was confirmed to the species level. In this study, additional morphometric data and SEM figures for these two stunt nematode were also provided. The representative sequences of rDNA fragment for these two species had the size of 1198bp were the first submitted to GenBank.
