In this study, five strains of Lentinus edodes from Taiwan, 4 wild strains and 6 cultivated strains from Japan were used in the di-mon mating method to breed a new strain for the cultivation in the low elevation areas in Taiwan. Ten single spore isolated each of the five wild strains from Taiwan were used as monokaryons and both wild and cultivated strains from Japan were used as dikaryons which resulted in 500 all possible crosses, isolates or di-mon matings. Twenty-four out of the 500 isolates produced primo rdia on agar medium with a strong vigor of mycelial growth. The cultivation trails of the 24 isolates at Kuan-tien (25 m from sea level) indicated that the isolate L107 (K53 x D54) was very promising because it produced an economic yield and the shape and size of its fruiting bodies were normal and large. From the large scale trials on different kinds of logs at different locations in the areas of less than 250m from sea level, isolate L107 produced economic yield and the quality of the fruiting bodies is also good. Therefore, it can be either cultivated in the low elevation areas in Taiwan, or used as a breeding material for further improvement.
