筆者承蒙農委會經費補助,八十二年六月到九月赴美國研習植物基因轉殖的技術,經由美國農業部Beltsville農業研究中心(Beltsville Agricultural Research Center, United States Department of Agriculture)之安排,在花卉及苗圃作物研究組(Florist and Nursery Plants Research Unit)進行研習。研習期間主要以木本觀賞植物及菊花等園藝作物爲試驗材料,研習內容包括:(一)DNA分離萃取(二)植物基因轉殖系統:1.電泳基因轉殖法2.電穿孔基因轉殖法3.粒子槍基因轉殖法(三)組織化學染色法分析GUS活性。茲將此次研習之技術與心得整理成文,提供國內進行基因轉殖研究者之參考。
This study program was supported by a grant from the Council of Agriculture, Executive Yuan, Republic of China. The author went to Florist and Nursery Plants Research Unit, Beltsville Agricultural Research Center, United States Department of Agriculture to study gene transformation techniques from June to September, 1993, Daring this period, woody ornamental plants and chrysanthemum were used as testing materials. The program included DNA isolation, gene transformation via electrophoresis, electroporation, particle gun; and histochemical staining f or GUS activity assay. This report describes various methods of gene transformation used in Florist and Nursery Plants Research Unit, BARC-USDA.
