本研究利用Tacora、Ansofie及Fame等三品種非洲菊,分別萃取其花蕾期 (開始轉色期)、半開期及全開期之植物組織DNA及RNA,針對控制進入類黃素代謝途徑、控制黃酮 (flavones) 合成和控制進入花青素合成等三個酵素基囚設計增幅用引子 (primers),利用聚合酵素鏈鎖反應 (PCR) 分析不同品種非洲菊之花色基因表現情形。結果顯示,測試之三個非洲菊品種DNA均可獲得此三種酵素基因之增幅產物。另外以反轉錄聚合酵素鏈鎖反應(RT-PCR)結果得知,種酵素基因在不同品種間及不同生長時期的表現並不相同。以chs而言,在半開期的Fame品種表現較明顯,而Tacora和Ansofie品種則提早在花蕾期表現。僅明顯表現在Fame品種之全開期,在Tacora、Ansofie品種中則並未表現。至於dfr皆以半開期之表現最為明顯。
Three pigment biosynthetic genes, chs (chalcone synthase), fnsll (flavone synthas2 Il), and dfr (dihydroflavonol 4-reductase), of Gerbera hybrida were analyzed in this study. The differences of gene expression among three different cultivars of Gerbera, including Tacora, Ansofic, and Fame, were assayed based on RT-PCR. The expression of chs, fnsll, and dfr genes varied on floral organ of different cultivars during inflorescence development. The highest level of chs expression was found in the pigmentation Stage of cultivar 'Fame', and at closed buds of cultivars 'Tacora' and 'Ansofie'. The fnsll was only expressed at fully opened inflorescence of 'Fame'. There was a uniform expression pattem of dfr among three Gerbera cultivars.
