玉米蛋白質品質改良工作中,為選拔含高離氨酸之玉米品系,必須有適當而快速的方法測定離氨酸 (lysine) 含量。本研究系改進 dye-binding capacity (DBC) 測定粗蛋白質方法,以 Udy colorimeter 測定玉米離氨酸含量。用 DBC 法所測離氨酸含量與氨基酸分析儀所測離氨酸之含量間之相關係數為 +0.96,但如未除去樣品中之脂肪,則相關係數減為 +0.38。
One problem in screening high-lysine genotypes of cereal crops is the determination of lysine content. The traditional method with amino acid analyzer is accurate but time-consuming and rather expensive in cost. Therefore, the quick and less-expensive dye-binding method was evaluated for its accuracy. Hybrid corn varieties Tainan No. 5 and Tainan No. 11 and No. 13 double cross opaque-2 lines were used for comparing the two methods of lysine determination.
