本試驗之目的在建立藥用植物黨參利用組織培養大量繁殖之方法。結果發現以黨參之葉、莖段、節、莖頂及根為不同來源之培植體誘導不定芽之形成，以接近莖頂部份之莖段最好、莖頂次之，再其次為節，而以葉部最差，而以根為培植體者，則因消毒不全導致發霉而無結果。以莖段為培植體則以距離莖頂5 cm以内之莖段，誘導不定芽能力最高(0.5 cm之莖段平均可獲得11個不定芽），其次為距莖5.1〜10 cm者，而以距莖頂10.1〜20 cm者較差；莖段誘導不定芽的產生，以含0.1 mg/1 NAA及2-4 mg/1 BA之1 BA培養基較佳。誘導不定芽之發根則以含有1〜2 mg/1 NAA，5%Sucrose ,及1/4 MS基礎鹽類的培養基最好。將含有良好根系之試管苗移植於含有I/3腐質土之壤土，或壤土：蛭石：腐質土＝2：1：1之介質中，成活率均可達100%。利用本研究所建立之培養模式，可大量繁殖黨參供藥用。
The purpose of this study is to establish the tissue culture techniques for mass propagation of a Chinese medicinal herb, Codonopsis radix. It was found that adventitious buds could be regenerated from the excised explant directly. Difference in the ability of forming adventitious buds was observed among different plant organs with stem segment near shoot tip showing the highest regeneration ability followed by shoot tip, node and leaf blade. Data were not obtained from root explant due to contamination. Apical part of the stem segment (within 5 cm from the shoot tip) produced higher adventitious buds when compared to basal part of the stem segment. Apical part of the stem segments could produce 11 adventitious buds on a medium containing MS salts with 3%sucrose, 0.9% agar, 0.1 mg/1 NAA and 2-4 mg/1 BA. Concentration of MS salts, NAA and sucrose affected not only root formation from adventitious buds, but also root number and length. The best combination was 1/4 strength of MS salts with 5% sucrose and 1-2 mg/1 NAA. The high survival rate of 100 % was recorded when plantlets were transplanted from test tube onto soil/peat moss (2:1), or soil/vermiculite/peat moss (2:1:1). Thus we concluded that tissue culture technique appears to possess potential for mass propagation to Codonopsis radix plant for commercial use.
